Target Information

Cortactin (Cttn) is a ubiquitous actin-binding protein originally identified as a substrate for Src. It contributes to actin cytoskeleton organization and cell shape, playing key roles in lamellipodia formation and cell migration. Cortactin accumulates in actin-enriched peripheral structures, facilitating actin network formation. It contains four major domains: N-terminal acidic (NTA), tandem repeats, proline-rich, and SH3 domains. The NTA domain associates with Arp2/3 and WASP complexes at F-actin branches. Cortactin promotes cell motility and invasion and is critical for invadopodia formation, where actin-rich protrusions degrade ECM in cancer cells. Phosphorylation occurs at Y421, Y466, Y482 by Src kinases and at S405, S418 by Erk kinases.
