Invitrogen™ BLOCK-iT™ Adenoviral RNAi Expression System

The pAd/BLOCK-iT™-DEST RNAi Gateway™ Vector enables efficient delivery and transient expression of short hairpin RNA (shRNA) in vivo using an adenoviral vector.
A novel cloning process inserts an approximately 50-bp DNA oligonucleotide immediately downstream of the U6 pol III promoter in the BLOCK-iT™ U6 entry vector. After recombination of the entry vector into the pAd/BLOCK-iT™-DEST vector, followed by viral production and transduction, the U6 promoter-driven shRNA can be transiently expressed in most dividing and non-dividing mammalian cell types.
The generated shRNA avoids host defense mechanisms and effectively induces RNAi-mediated gene knockdown.
