Product Specific Information
The eBio124-1D1 monoclonal antibody reacts with human CD7, also known as gp40 or Leu9. CD7 is a 40 kDa receptor and a member of the immunoglobulin gene superfamily.
The N-terminal amino acid sequence (aa1–107) is highly homologous to Ig kappa light chain sequence, while the carboxyl-terminal region is proline-rich and may form a stalk projecting the Ig domain.
CD7 is expressed on:
- Most immature and mature T lymphocytes
- T cell leukemias
- Natural killer (NK) cells
- A small subset of normal and malignant B cells
Functional Role:
Cross-linking CD7 modulates T cell and NK cell activity, inducing calcium flux, adhesion molecule expression, cytokine secretion, and proliferation. CD7 directly associates with phosphoinositol 3′-kinase, and its ligation induces D-3 phosphoinositide production and tyrosine phosphorylation.
A clonogenic subpopulation of CD34(+) CD38(-) cord blood cells expressing CD45RA, HLA-DR, and high levels of CD7 has lymphoid lineage potential, generating B cells, NK cells, and dendritic cells but not myeloid or erythroid cells.
The CD7(+) phenotype distinguishes primitive human lymphoid progenitors from pluripotent stem cells.
It has also been suggested that CD7 cooperates with CD28 during Treg function; mice lacking both CD28 and CD7 show reduced Treg numbers and suppressive activity.
Applications Reported: Flow cytometric analysis
Applications Tested: Flow cytometry of normal human peripheral blood cells (≤1 µg/test).
A test is defined as the amount of antibody staining cells in 100 µL volume (10⁵–10⁸ cells/test). Antibody titration is recommended for optimal performance.
Target Information
