Target Information

PROZ (Protein Z) is a vitamin K-dependent plasma glycoprotein, approximately 62 kDa and 396 amino acids in length. It contains four domains: a gla-rich region, two EGF-like domains, and a trypsin-like domain. Although structurally related to serine proteases such as factors VII, IX, X, and protein C, PROZ lacks catalytic activity due to the absence of a key serine residue. It plays a role in the coagulation cascade by facilitating the degradation of factor Xa through protein Z-related protease inhibitor (ZPI), enhancing the reaction rate up to 1000-fold. Deficiency states have been linked to both thrombosis and bleeding tendencies, though mechanisms remain unclear.
