Target Information
Bassoon은 약 420 kDa 크기의 단백질로, 시냅스 전 말단(presynaptic nerve terminal)에 위치하며 시냅스 소포 순환의 구조적 및 기능적 조직화에 관여하는 것으로 알려져 있습니다.
Rat과 Mouse에서 각각 96%의 서열 상동성을 보이며, 인간 BASSOON 유전자는 최근 클로닝 및 위치 확인이 완료되었습니다.
Bassoon은 두 개의 double-zinc finger, 세 개의 coiled-coil 영역, 두 개의 polyglutamine 도메인을 포함하는 것으로 예측됩니다.
C-말단의 polyglutamine 도메인은 일부 인간 단백질(예: Huntingtin)에서 비정상적 확장이 신경퇴행 질환을 유발할 수 있는 것으로 알려져 있어 주목받고 있습니다.
Bassoon은 시냅스 말단에서 postsynaptic density 반대편에 집중되어 있으며, 배양된 뉴런에서는 GABA(A) 및 GluR1 수용체와 공위치(colocalization)하는 것으로 관찰됩니다.
또한 Piccolo 단백질과도 공위치하여 시냅스 접합부에서 세포골격 구조 유지에 관여할 가능성이 제시되었습니다.
