Product Specific Information
- Recommended positive control: 293T cells
- For IHC (paraffin), perform heat-mediated antigen retrieval (0.1M citrate buffer at 95°C for 20 minutes).
- To avoid nonspecific binding, incubate in normal rabbit anti-mouse serum.
- Incubate primary antibody overnight and use biotin-streptavidin amplification kit.
- Block endogenous peroxidase with 0.3% H₂O₂ for 30 minutes.
Target Information
BRCA1 (breast and ovarian cancer susceptibility protein 1) is a nuclear phosphoprotein involved in maintaining genomic stability and acts as a tumor suppressor. It forms part of the BRCA1-associated genome surveillance complex (BASC), interacting with DNA repair and transcription machinery. Mutations in BRCA1 account for ~40% of inherited breast cancers and over 80% of inherited breast and ovarian cancers.
Alternative splicing affects its localization and function, with several disease-associated variants identified. A related pseudogene also exists on chromosome 17.
