After incubation with the primary antibody, slides were incubated with biotinylated secondary antibody, followed by alkaline phosphatase-streptavidin and chromogen.
Target Information
AHR (Ah Receptor) belongs to a family of proteins including its dimerization partner ARNT (HIF-1 Beta) and Drosophila proteins PER and SIM.
AHR contains an N-terminal PAS domain (~200 amino acids) and binds to specific DNA enhancer sequences to initiate transcription of cytochrome P-450 (CYP1A1) gene.
It is a ligand-activated transcription factor regulating biological responses to planar aromatic hydrocarbons and xenobiotic-metabolizing enzymes such as cytochrome P450.
Known ligands include polycyclic and halogenated aromatic compounds such as 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD).
In non-ligand activated cells, AHR is complexed with HSP90 in the cytoplasm. Upon ligand binding, AHR translocates to the nucleus, dissociates from HSP90, and forms a heterodimer with ARNT.
Diseases associated with AHR include eosinophilic fasciitis and seborrheic dermatitis.
