Product Specific Information
For IHC(P), use heat-induced antigen retrieval in pH 6.0 citrate buffer. After incubation with the primary antibody, slides were treated with biotinylated secondary antibody, followed by alkaline phosphatase-streptavidin and chromogen.

Target Information
AHR (Aryl Hydrocarbon Receptor) is part of a protein family that includes ARNT (HIF-1 Beta) and Drosophila proteins PER and SIM. It contains an N-terminal PAS domain (~200 amino acids) and is found in various tissues. AHR binds specific DNA enhancer sequences to initiate transcription of cytochrome P-450 (CYPIA1) mRNA.
AHR encodes a ligand-activated transcription factor regulating biological responses to planar aromatic hydrocarbons and xenobiotic-metabolizing enzymes such as cytochrome P450. Known ligands include polycyclic and halogenated aromatic compounds like TCDD. Unlike steroid or thyroid hormone receptors, no physiological ligand is known.
In non-ligand activated cells, AHR complexes with HSP90 in the cytoplasm. Upon agonist binding, AHR transforms into a nuclear DNA-binding form, dissociates from HSP90, and forms a heterodimer with Arnt (AHR nuclear translocator). Diseases associated with AHR include eosinophilic fasciitis and seborrheic dermatitis.
