Target Information
AHR (Ah Receptor) belongs to a family of proteins that includes its dimerization partner ARNT (HIF-1 Beta) and the Drosophila proteins PER and SIM.
AHR contains an N-terminal PAS domain (~200 amino acids) and binds specific DNA enhancer sequences to initiate transcription of cytochrome P-450 (CYPIA1) mRNA.
It acts as a ligand-activated transcription factor regulating biological responses to planar aromatic hydrocarbons.
Known ligands include polycyclic and halogenated aromatic compounds such as 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD).
In non-ligand activated cells, AHR is complexed with HSP90 in the cytoplasm. Upon ligand binding, AHR transforms into a nuclear DNA-binding form, dissociates from HSP90, and forms a heterodimer with Arnt.
Diseases associated with AHR include eosinophilic fasciitis and seborrheic dermatitis.
