Product Specific Information

In Western blot, multiple bands may be seen at 100–120 kDa representing post-translational modification of HIF-1α.
For WB, testing on nuclear extracts is recommended.


Target Information

HIF1-alpha (HIF1A) is a subunit of HIF1, a transcription factor found in mammalian cells under reduced oxygen tension. HIF-1 is a heterodimer consisting of alpha and beta subunits, both belonging to the basic-helix-loop-helix PAS family of transcription factors.

HIF1 functions as a transcriptional regulator of the adaptive response to hypoxia, activating transcription of genes that increase oxygen delivery or facilitate metabolic adaptation. These include erythropoietin, glucose transporters, glycolytic enzymes, VEGF, and others.

HIF1-alpha regulates hypoxia-mediated apoptosis, cell proliferation, and tumor angiogenesis. It interacts with p53 under hypoxic conditions, influencing ubiquitination and degradation. Induction of NOX4 by HIF1-alpha contributes to ROS maintenance and proliferation after hypoxia.

In humans, HIF1A is located on chromosome 14q and binds to p300, forming complexes that activate hypoxia-responsive genes. Hypoxia plays a key role in diseases such as ischemia, cancer, pulmonary hypertension, and COPD.
