Product Specific Information

The eBioOMAK-D antibody reacts with mouse perforin (pore-forming protein, pfp, Prf).
Perforin is a cytolytic mediator found in cytotoxic T lymphocytes (CTL) and natural killer (NK) cells, playing a key role in immune defense against tumors and viral infections.

• Recognizes a ~70 kDa band in lysates of CTLL-2 mouse cytotoxic cell line and IL-2 stimulated mouse splenocytes.
• Staining increases upon stimulation with IL-2 or anti-CD3/CD28.
• Perforin Knock-out splenocytes do not stain with eBioOMAK-D nor show protein detection by western blot.
• In IL-2 stimulated mouse splenocytes, NK cells (CD49b+) contain perforin, while CD8 cells show variable expression depending on culture conditions.
• Cross-reactive to human perforin; co-stains CD56+ PBMC cells.
• Granzyme B expression precedes and exceeds perforin expression during stimulation.

For intracellular staining and flow cytometry, use the Foxp3 buffer system (Product #00-5523).
Other buffers may yield varying results.

Applications Tested: Flow cytometric analysis of stimulated mouse splenocytes using the Foxp3/Transcription Factor Staining Buffer Set.

Target Information

Perforin is a major cytolytic protein stored and released by cytotoxic granules. It plays a critical role in immune defense against tumors and viral infections mediated by cytotoxic lymphocytes.
Perforin is a 555 amino acid protein (~70–75 kDa) with a 21 amino acid signal peptide and shares homology with complement component C9.
Expressed only in killer cell lines, not in helper T cells or other tumor cells.
Defects in the perforin gene cause familial hemophagocytic lymphohistiocytosis type 2 (HPLH2), a rare autosomal recessive disorder of childhood.
Alternative splicing results in multiple transcript variants.
