Preparation:
Produced from sera of rabbits immunized with highly pure Recombinant Human Amphiregulin. The Anti-Human Amphiregulin-specific antibody was purified by affinity chromatography using an immobilized Human Amphiregulin matrix.

Sandwich ELISA:
To detect Human Amphiregulin by sandwich ELISA (100 µL/well), use 0.5–2.0 µg/mL of this antibody. When paired with PeproTech Biotinylated Anti-Human Amphiregulin (500-P322Bt) as a detection antibody, it enables detection of 2000–4000 pg/mL of Recombinant Human Amphiregulin.

Western Blot:
For Western blot detection of Human Amphiregulin, use 0.1–0.2 µg/mL. With compatible secondary reagents, the detection limit is 1.5–3.0 ng/lane under reducing or non-reducing conditions.

Target Information

Amphiregulin (AR) binds to the EGF receptor (EGFR) with lower affinity than EGF. The mature secreted AR is an 84-amino acid glycosylated growth regulator derived from a 252-amino acid transmembrane precursor.
Seven ligands—EGF, TGFα, AR, HB-EGF, cripto-1, epiregulin, and betacellulin—bind to the extracellular domain of EGFR. Each contains an EGF-like domain with six cysteine residues forming three disulfide-bonded loops.
AR protein is localized in epithelial tissues of the colon, stomach, pancreas, breast, and placenta, and is overexpressed in several human cancers including breast, colon, stomach, and pancreas.
