Product Specific Information
1:1,000 희석의 MA5-45081은 2 µg의 정제된 poly-ubiquitin 체인(Ub1–Ub7, K63-linked, K48-linked)에서 Ubiquitin 검출에 충분함. ECL immunoblot 분석 시 goat anti-rabbit IgG:HRP를 2차 항체로 사용함.
이 항체는 ubiquitinated 단백질(단일 및 다중 ubiquitinylated, K29-, K48-, K63-linked)을 검출하며, free ubiquitin은 인식하지 않음.
Target Information
Ubiquitin은 세포 내 단백질을 26S proteasome을 통해 분해 대상으로 지정하는 highly conserved 단백질임. 전구체 형태로 합성되며, polyubiquitin 체인 또는 다른 단백질과 결합된 형태로 존재함.
이 유전자는 N-말단의 ubiquitin과 C-말단의 ribosomal protein S27a로 구성된 융합 단백질을 암호화함. 효모에서 발현 시 번역 후 처리되어 free ubiquitin과 ribosomal protein S27a가 생성됨. S27a는 40S ribosomal subunit 구성 요소로 cytoplasm에 존재하며 C4-type zinc finger domain을 포함함.
이 유전자는 다수의 pseudogene을 가지며, ribosomal protein L40과 S30도 각각 ubiquitin 또는 ubiquitin-like protein과 융합 형태로 합성됨. 여러 가지 alternative splicing 변형이 보고됨.
