Target Information
CD73 (Ecto-5′-nucleotidase) catalyzes the conversion of purine 5′-mononucleotides to nucleosides at neutral pH, with AMP as the preferred substrate.
It exists as a homodimer of two identical 70 kDa subunits anchored to the plasma membrane via a glycosyl phosphatidylinositol linkage.
CD73 serves as a marker of lymphocyte differentiation, and its deficiency is associated with various immunodeficiency disorders.
Other forms of 5′-nucleotidase exist in the cytoplasm and lysosomes, differing in substrate affinity, magnesium ion requirement, ATP activation, and phosphate inhibition.
The CD73 gene is localized to chromosome 6q14–q21. Mutations can lead to joint and arterial calcification and intestinal tuberculosis.
Two transcript variants encoding different isoforms have been identified.
