Target Information
The mammalian thioredoxin reductases (TrxRs) are a family of selenocysteine-containing pyridine nucleotide-disulfide oxidoreductases. Mammalian TrxRs are homologous to glutathione reductase with respect to primary structure, including the conserved redox catalytic site (-Cys-Val-Asn-Val-Gly-Cys-), but distinctively possess a C-terminal extension with a catalytically active penultimate selenocysteine (SeCys) residue (-Gly-Cys-SeCys-Gly).
TrxR is a homodimeric protein containing an FAD prosthetic group, NADPH binding site, and redox catalytic site. Electrons are transferred from NADPH via FAD and the active-site disulfide to the C-terminal SeCys-containing redox center, which reduces substrates like thioredoxin.
TrxR family members (TrxR1, TrxR2, TrxR3) are 55–58 kDa proteins involved in cellular protection against oxidative stress and regulation of transcription factors and various biological processes.
