Target Information

RNA polymerase (Pol) III synthesizes tRNA, 5S rRNA, 7SL RNA, and U6 snRNA and is overexpressed in many transformed cell lines and tumors. To maintain rapid growth, cells must produce high levels of Pol III–transcribed RNA, requiring TFIIIB and TFIIIC2 complexes.
The TFIIIC2 complex consists of five subunits—TFIIIC220, TFIIIC110, TFIIIC102, TFIIIC90, and TFIIIC63—overexpressed in malignant cells such as ovarian carcinomas. TFIIIC2 recruits RNA Pol III and TFIIIB to promoter elements and may play a key role in deregulation of cancer cells.
TFIIIB includes TBP, BRF1, and TFIIIB subunits, which are also upregulated in transformed cells. Tumor suppressors RB and p53, often inactivated in carcinomas, lose their ability to bind and regulate TFIIIB function.
