Reconstitution:
Briefly centrifuge the vial before opening to bring contents to the bottom. Reconstitute lyophilized rat IL-10 in 5 mM Tris, pH 7.2, to a concentration of 0.1–1.0 mg/mL. Further dilution should be made in medium or buffered solution containing carrier protein (e.g., PBS with 0.1% BSA).

Storage:
Lyophilized preparation is stable at 2–8°C, but should be kept at -20°C for long-term storage, preferably desiccated. After reconstitution, stable for up to one week at 2–8°C. For maximal stability, divide into aliquots and store at -20°C. Avoid repeated freeze/thaw cycles.

Target Information

IL-10 encodes a cytokine primarily produced by monocytes and lymphocytes. It plays a key role in immunoregulation and inflammation by down-regulating Th1 cytokines, MHC class II antigens, and costimulatory molecules on macrophages. It enhances B cell survival, proliferation, and antibody production. IL-10 also blocks NF-kappa B activity and regulates the JAK-STAT signaling pathway. Knockout studies indicate its essential role in intestinal immune regulation. Mutations in IL10 are linked to increased susceptibility to HIV-1 infection and rheumatoid arthritis. Associated diseases include Graft-Versus-Host Disease and Human Immunodeficiency Virus Type 1.
