Description:
The 4B4 (4B4-1) monoclonal antibody reacts with human CD137 (4-1BB, TNFRSF9), an inducible member of the TNFR family expressed on T cells, NK cells, dendritic cells, granulocytes, and mast cells. CD137 signaling is involved in T cell activation, survival of activated and CD8 memory T cells, and suppression of myelopoiesis and dendritic cell development. Stimulation of CD137 promotes expansion of CD4⁺CD25⁺ T regulatory cells ex vivo. Its ligand, 4-1BBL, is expressed on activated macrophages, mature B cells, hematopoietic stem cells, and myeloid progenitor cells.

Applications Tested:
Tested on two-day Con A-stimulated normal human peripheral blood cells. Recommended use: 5 µL (0.125 µg) per test in 100 µL final volume. Cell number may range from 10⁵ to 10⁸ cells/test. Antibody titration is recommended for optimal performance.

Light Sensitivity:
This tandem dye is sensitive to photo-induced oxidation. Protect from light.

Fixation:
Samples can be stored in IC Fixation Buffer (Product #00-8222) or 1-step Fix/Lyse Solution (Product #00-5333-54) for up to 3 days at 4°C in the dark with minimal impact on brightness and FRET efficiency.

Target Information

CD137 (TNFRSF9, 4-1BB) is an inducible costimulatory molecule expressed mainly on activated T cells. Its ligand 4-1BBL is found on activated macrophages, mature B cells, hematopoietic stem cells, and myeloid progenitor cells. CD137 signaling maintains activated and memory T cell survival, promotes T cell clonal expansion, and suppresses myelopoiesis and dendritic cell development. Soluble CD137 may act as a negative regulator of immune responses.
