Product Specific Information
This antibody is specific for human SIAH2 and Drosophila SINA in proliferating cells and stem cells. It does not detect Drosophila SINAH or normal, non-dividing cells.
Target Information
SIAH-2 (seven in absentia homolog 2) is an E3 ubiquitin ligase involved in proteasome-mediated degradation of protein substrates. The SIAH-2 gene encodes a 324 amino acid protein sharing 77% identity with human SIAH-1 and 68% with Drosophila sina, which is essential for R7 photoreceptor development.
SIAH-2 targets TRAF2 for degradation under stress conditions such as hypoxia and regulates HIF-1α prolyl hydroxylase 3 (PHD3) degradation, correlating with increased SIAH-2 transcription. It can also decrease TNF-α-dependent induction of JNK activity and NFκB activation.
SIAH-2-deficient mice exposed to hypoxia exhibit impaired respiratory response and reduced hemoglobin levels.
