Target Information
Scn5a is an integral membrane protein and tetrodotoxin-resistant voltage-gated sodium channel subunit. It is primarily expressed in cardiac muscle and responsible for the initial upstroke of the action potential in an electrocardiogram.
Defects in Scn5a cause Long QT Syndrome type 3 (LQT3), an autosomal dominant cardiac disease. Alternative splicing generates several transcript variants encoding different isoforms.
Voltage-gated sodium channels (VGSCs) mediate regenerative membrane depolarization and electrical signal conduction in nerves and muscles. Scn5a expression is also detected in lymphocytes, glia, and fibroblasts.
Functionally, Scn5a mediates voltage-dependent Na⁺ ion permeability, forming sodium-selective channels that open or close in response to membrane voltage changes. Channel inactivation is regulated by intracellular calcium levels.
Diseases associated with SCN5A include Long QT Syndrome-3 and Brugada Syndrome 1.
