Product Specific Information

For IHC on PFA-fixed tissues, incubate the antibody for 30 minutes at room temperature. Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 minutes at 95°C, followed by cooling at room temperature for 20 minutes.

Target Information

CD30 (Ki-1, TNF Receptor Superfamily Member 8) is a type I transmembrane glycoprotein belonging to the TNF receptor superfamily. It was originally identified as a cell surface antigen of Hodgkin’s and Reed-Sternberg cells using monoclonal antibody Ki-1. Its ligand, CD30L (CD153), mediates diverse cellular responses including proliferation, differentiation, and apoptosis. CD30 plays a critical role in Hodgkin’s disease and other CD30+ lymphomas, and functions as a costimulatory molecule in thymic negative selection. It is expressed in Hodgkin’s and Reed-Sternberg cells, certain non-Hodgkin’s lymphomas (e.g., Burkitt’s lymphoma), virus-infected T and B cells, and activated normal T and B cells. Soluble CD30 (sCD30) serves as a marker of Th2 immune response. CD30 interacts with TRAF2 and TRAF5 to activate NF-κB signaling and positively regulate apoptosis, limiting autoreactive CD8 T cell proliferation. Two alternatively spliced transcript variants encode distinct isoforms. CD30 is expressed by mononuclear cells in Hodgkin’s lymphoma, Reed-Sternberg cells, most Anaplastic Large Cell Lymphomas (ALCL), and embryonal carcinomas.
