• For PFA-fixed tissues: incubate antibody for 30 minutes at room temperature.
• For formalin-fixed tissues: heat sections in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 minutes at 95°C, then cool at room temperature for 20 minutes.

Target Information

CD30 (Ki-1, TNF Receptor Superfamily Member 8) is a type I transmembrane glycoprotein belonging to the TNF receptor superfamily.
Originally identified on Hodgkin’s and Reed-Sternberg cells using monoclonal antibody Ki-1, CD30 binds to its ligand CD30L (CD153) to mediate diverse cellular responses including proliferation, activation, differentiation, and apoptosis.

CD30 plays a critical role in the pathophysiology of Hodgkin’s disease and other CD30⁺ lymphomas. It also functions as a costimulatory molecule in thymic negative selection and is expressed on activated T and B cells, some non-Hodgkin’s lymphomas, and virus-infected lymphocytes.

The soluble form (sCD30) serves as a marker for Th2 immune response. TRAF2 and TRAF5 interact with CD30 to activate NF-κB signaling. CD30 positively regulates apoptosis and helps control autoreactive CD8⁺ T cell proliferation, contributing to immune homeostasis. Two alternatively spliced transcript variants encoding distinct isoforms have been reported.
