Target Information

DNA methylation, the addition of methyl groups to cytosine bases in CpG dinucleotides, is crucial for development and gene regulation. The methylation pattern results from enzymatic methylation and demethylation processes. MBD4, also known as MED1, is a mammalian demethylase associated with the methyl-CpG-binding domain (MBD). It catalyzes the removal of methyl groups from methylated cytosine, reverting them to cytosine.

Proteins such as MeCP2 and MBD1 repress transcription by binding to methylated DNA. MBD2 and MBD4 colocalize with heavily methylated satellite DNA, mediating methylation-related biological functions. MBD3, however, does not bind methylated DNA. MBD family members are expressed in somatic tissues, with MBD1 and MBD2 expression reduced in embryonic stem cells. MBD4 shares homology with bacterial DNA N-glycosylases/lyases and can be mutated in microsatellite unstable tumors.
