Target Information

The tumor suppressor protein p53 is a sequence-specific transcription factor activated by cellular stress. It mediates cell cycle arrest or apoptosis in response to DNA damage or pyrimidine nucleotide starvation. p53 activation leads to transcription of genes such as p21waf1, regulating G1 or G2/M arrest.

The structure of p53 includes:

  • N-terminal transactivation domain
  • Central DNA-binding domain
  • Oligomerisation domain
  • C-terminal regulatory domain

Phosphorylation at Ser15 is essential for activation and stabilization. Ser392 phosphorylation is associated with tumor formation. p53 plays a crucial role in preventing proliferation of damaged cells and acts as an anticancer agent. Elevated p53 levels are linked to aging, oxidative stress, and deficits in learning and memory.

Mutations in p53 disrupt DNA binding and tumor suppression, contributing to malignancy. Alterations of the TP53 gene occur as somatic and germline mutations, including those found in Li-Fraumeni syndrome.
