Target Information
SIRP alpha (CD172a, signal-regulatory protein alpha)는 수용체형 막관통 당단백질로, 과립구, 수지상세포(DC), 대식세포, 비만세포 및 조혈줄기세포 등 골수계 세포에서 발현됩니다.
SIRP alpha는 여러 활성화된 티로신 키나아제(EGFR, PDGFR, src, insulin receptor 등)의 기질로 작용하며, 수용체 티로신 키나아제 신호전달 경로의 음성 조절에 관여합니다.
CD47과의 결합은 SIRP alpha의 ITIM 영역에서 티로신 인산화 및 SHP-1, SHP-2의 활성화를 유도하며, 대식세포의 식세포 작용 억제 및 T 세포, DC의 활성화 조절에 관여합니다.
암세포는 CD47을 과발현하여 SIRP alpha를 활성화시키고, 대식세포에 의한 세포 파괴를 억제함으로써 종양 성장에 영향을 줍니다.
