Product Specific Information

The 61D3 monoclonal antibody reacts with human CD14, a 53–55 kDa GPI-linked glycoprotein expressed on monocytes, interfollicular macrophages, and some dendritic cells. CD14 binds LPS-LBP complexes with high affinity.

Each product includes:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and validated for human peripheral blood cells at 5 µL/test (0.6 µg/test). A test corresponds to staining a 100 µL sample containing 10⁵–10⁸ cells.

Note:
NovaFluor dyes are incompatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI). Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.

CellBlox Plus Blocking Buffer Usage:

• Add 5 µL per sample before mixing with cells.
• For single-color controls: 5 µL per 100 µL of cell sample (10³–10⁸ cells).
• Required for optimal staining with NovaFluor conjugates.

NovaFluor conjugates utilize Phiton™ technology, providing engineered fluorescent signatures optimized for minimal spillover and spectral spread.

Target Information

CD14 is a 55 kDa GPI-anchored glycoprotein expressed on monocytes, macrophages, and neutrophils. It functions as a lipopolysaccharide (LPS) receptor and exists in both membrane-bound and soluble forms (sCD14). CD14 interacts with LPS-binding protein (LBP) and plays roles in immune recognition, inflammation, and apoptosis. Dysregulation of CD14 is associated with infections such as Mycobacterium chelonae and conditions like croup.
