This antibody is directed against human Smad2 protein. BLAST analysis suggests cross-reactivity with Smad2 from human, mouse, and rat based on 100% sequence homology. No reactivity was detected with Smad1, Smad3, Smad4, or Smad7 in over-expressed cell lysates (Personal Communication, Kathleen Flanders, CCR-NCI, Bethesda, MD).
Target Information
SMAD2 (MADH2, MAD2) regulates multiple cellular processes such as proliferation, apoptosis, and differentiation.
It interacts with TGF-beta receptors via the SMAD anchor for receptor activation, a key step in TGF-beta signaling.
Phosphorylation of threonine 8 by ERK1 enhances SMAD2 transcriptional activity, which is negatively regulated by calmodulin.
Upon TGF-beta stimulation, SMAD2 is phosphorylated, dissociates from SARA, and associates with SMAD4 for nuclear translocation and transcriptional regulation.
SMAD2 can also mediate activin signaling and exists in alternatively spliced transcript variants.
