Product Specific Information
Reconstitution: Add 50 µL double distilled water (DDW), depending on sample size.
The antibody ships as a lyophilized powder at room temperature. Upon arrival, store at -20°C.
Reconstituted solution can be stored at 4°C for up to 1 week. For longer periods, store aliquots at -20°C.
Avoid multiple freeze/thaw cycles. Centrifuge all antibody preparations before use (10,000 × g, 5 min).
Target Information
Inositol 1,4,5-trisphosphate (IP3) acts as a second messenger for various growth factors, hormones, and neurotransmitters.
Upon binding to IP3R, IP3 triggers intracellular calcium release from luminal stores to the cytosol.
Functional IP3R is a homo- or heterotetramer (~240 kDa glycoprotein subunits) structurally and functionally related to the ryanodine receptor.
Both receptors interact with immunophilin FKBP12.
Phosphorylation of IP3R by PKC enhances calcium release; dephosphorylation by calcineurin reduces release, causing calcium oscillations.
Mammalian IP3R subunits are encoded by three distinct genes (IP3R-I, IP3R-II, IP3R-III) with differential tissue expression:
- IP3R-I: heart, liver, kidney, ovary, cerebellar Purkinje neurons
- IP3R-II: predominantly brain
- IP3R-III: pancreatic islets, kidney, gastrointestinal tract
