Target Information
Amyloid beta peptide (Abeta/Beta-amyloid)는 알츠하이머병 환자의 뇌에서 발견되는 아밀로이드 플라크의 주요 구성 성분입니다. Abeta 펩타이드는 40–43개의 아미노산으로 구성되어 있으며, beta-amyloid precursor protein (beta APP)에서 두 단계의 절단 과정을 통해 생성됩니다.
첫 번째 단계에서는 beta APP의 아미노 말단 도메인이 beta-secretase (BACE)에 의해 절단되며, 이후 gamma-secretase가 막 내 절단을 수행하여 amyloid beta peptide를 생성합니다.
Gamma-secretase는 presenilin-1 및 presenilin-2를 중심으로 하는 다단백 복합체로 알려져 있으며, nicastrin과 결합하여 amyloid beta 생성 조절에 관여합니다.
Abeta는 세포외 섬유성 단백질로서 아밀로이드 중심, 신경돌기 플라크, 신경섬유 엉킴(neurofibrillary tangles) 등에서 발견됩니다.
알츠하이머병은 이러한 비정상 단백질 침착으로 특징지어지는 대표적인 노인성 치매 질환이며, 베타 아밀로이드 침착은 루이체 치매, 다운 증후군, Dutch형 아밀로이드증, 뇌혈관 아밀로이드증 등에서도 관찰됩니다.
