Target Information
Pax 유전자군은 배아 발달 초기 특정 조직에서 활성화되는 전사 인자(transcription factor)로 구성됩니다. Pax 단백질은 paired box가 암호화하는 DNA 결합 도메인을 포함하며, 유전적 프로그램을 실행할 수 있습니다.
여러 Pax 유전자는 발달 관련 돌연변이와 연관되어 있으며, 예를 들어 Pax-3은 Waardenburg 증후군, Pax-6은 Aniridia, Pax-2는 Wilms 종양과 관련이 있습니다.
배아 발생 중 Pax-2는 신장 발달 과정에서 발현되며, 특히 metanephric mesenchyme의 응축 및 초기 상피 구조에서 발현됩니다. 그러나 성숙한 세뇨관 상피에서는 Pax-2 mRNA와 단백질 수준이 급격히 감소합니다.
Wilms 종양의 미분화된 상피에서는 Pax-2 발현이 지속되며, 이는 poorly differentiated epithelial cell의 증식과 관련됩니다.
또한 WT1 종양 억제 단백질의 발현은 Pax-2의 하향 조절과 일치하며, WT1이 Pax-2 전사를 직접 억제할 수 있음을 시사합니다.
