Description:
The WM-53 monoclonal antibody reacts with human CD33 (GP67, P67), a 67 kDa type I transmembrane glycoprotein belonging to the Siglec (sialic acid-binding Ig superfamily lectin) family. It is specific to the hematopoietic compartment and expressed on monocytes, activated T cells, granulocytes, myeloid progenitors, and mast cells.

Applications Reported:
The WM-53 (WM53) antibody is reported for use in flow cytometric analysis.

Applications Tested:
Pre-titrated and tested by flow cytometric analysis of normal human lysed whole blood. Use at 5 µL (0.5 µg) per test. A test is defined as the amount of antibody that will stain a cell sample in a final volume of 100 µL. Cell number may range from 10⁵ to 10⁸ cells/test.

Target Information

CD33 is a transmembrane protein of the sialic acid-binding immunoglobulin-like lectin (Siglec) family. It contains immunoreceptor tyrosine-based inhibitory motifs (ITIMs) that recruit protein tyrosine phosphatases SHP-1 and SHP-2, and mediate receptor internalization.
It is expressed on myelomonocytic lineage cells, binds sialic acid residues on N- and O-glycans, and serves as a therapeutic target for acute myeloid leukemia.
CD33 is found on granulocyte and macrophage precursors in bone marrow, but not on pluripotent stem cells. It is also a marker for peripheral monocytes and is useful for distinguishing myelogenous leukemia cells from lymphoid or erythroid leukemias.
Diseases associated with CD33 dysfunction include gallbladder lymphoma and extracutaneous mastocytoma.
