Product Specific Information

The O323 monoclonal antibody reacts with human CD27, a lymphocyte-specific member of the TNFR superfamily. CD27 is expressed by a subset of thymocytes and virtually all mature T cells, and is upregulated upon T-cell stimulation. CD27 binds to CD70, playing an important role in T cell–B cell interaction.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-titrated and tested on normal human peripheral blood cells.
Recommended usage: 5 µL (1 µg) per test in 100 µL final volume. Cell number range: 10⁵–10⁸ cells/test.

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains with NovaFluor dyes.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.
  • Add 5 µL per sample before combining with cells.
  • For single-color controls, use 5 µL buffer per 100 µL of cell sample.

Target Information

CD27 is a 50 kDa member of the TNF receptor superfamily (including CD40 and CD30). It regulates cell proliferation and death and is primarily expressed in lymphocytes. CD27 forms disulfide-linked homodimers on mature thymocytes, peripheral blood T cells, and some B cells. Activation of T cells increases CD27 expression and releases soluble CD27 (28–32 kDa), a prognostic marker for B cell malignancies.

CD27 interacts with CD70 and other proteins such as SIVA, influencing apoptosis and T cell activation. Dysfunction is associated with Lymphoproliferative Syndrome 2 and Autosomal Recessive Lymphoproliferative Syndrome.
