Product Specific Information

The O323 monoclonal antibody reacts with human CD27, a lymphocyte-specific member of the TNFR superfamily. CD27 is expressed by a subset of thymocytes and most mature T cells, and is upregulated upon T-cell stimulation. It binds to CD70 and plays an important role in T cell–B cell interaction.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and tested by flow cytometry using normal human peripheral blood cells at 5 µL (1 µg) per test.
A test is defined as the amount of antibody sufficient to stain a cell sample in a final volume of 100 µL. Cell number may range from 10⁵ to 10⁸ cells/test.

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains with NovaFluor dyes.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.
  • Add 5 µL per sample to antibody cocktails before combining with cells.
  • For single-color controls, add 5 µL per 100 µL of cell sample (10³–10⁸ cells).

NovaFluor conjugates utilize Phiton™ technology, based on engineered nucleic acid dye structures for optimized fluorescent signatures minimizing spillover and spread.

Target Information

CD27 is a 50 kDa member of the TNF receptor superfamily, including CD40 and CD30. It regulates cell proliferation and death. CD27 and its ligand CD70 are primarily expressed in lymphocytes. CD27 forms disulfide-linked homodimers on mature thymocytes, peripheral blood T cells, and a subset of B cells. Activation via the TCR-CD3 complex upregulates CD27 expression and induces release of its soluble form (sCD27), a prognostic marker in B cell malignancies. CD27 also interacts with SIVA, a proapoptotic protein, influencing apoptosis pathways.
