Product Specific Information

The O323 monoclonal antibody reacts with human CD27, a lymphocyte-specific member of the TNFR superfamily. CD27 is expressed by thymocytes and mature T cells and is upregulated upon T-cell stimulation. It binds to CD70 and plays an important role in T cell-B cell interactions.

Each product contains:

  • 1 vial of NovaFluor conjugate
  • 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-titrated and tested on normal human peripheral blood cells. Recommended use at 5 µL (0.8 µg) per test in a final volume of 100 µL.

Important Notes:

  • NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
  • Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for best results.
  • CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) must be used for optimal staining. Add 5 µL per sample regardless of NovaFluor count.
  • For single-color controls, use 5 µL of blocking buffer per 100 µL of cell sample (10³–10⁸ cells).

NovaFluor conjugates utilize Phiton™ technology, enabling engineered fluorescent signatures with minimized spectral spillover and spread.

Target Information

CD27 is a 50 kDa member of the TNF receptor superfamily (including CD40 and CD30). It regulates cell proliferation and death and is primarily expressed on lymphocytes. CD27 forms a disulfide-linked homodimer on thymocytes, peripheral T cells, and some B cells. Activation via TCR-CD3 increases CD27 expression and releases soluble CD27 (sCD27), a prognostic marker for B cell malignancies. CD27 dysfunction is associated with Lymphoproliferative Syndrome 2 and Autosomal Recessive Lymphoproliferative Syndrome.
