Product Specific Information

The MTS510 monoclonal antibody reacts with the mouse Toll-like receptor 4 (TLR4)/MD-2 complex. TLR4 is a type I transmembrane protein involved in innate immunity and inflammation, forming a complex with MD-2 and CD14 for LPS recognition and signaling.
In mice, TLR4 is expressed by thioglycolate-elicited peritoneal macrophages and is downregulated upon LPS stimulation. The TLR4 gene is defective in C3H/HeJ and C57BL/10ScCr mice, known as hyporesponders to LPS.

MTS510 co-immunoprecipitates MD-2 (~30 kDa) and TLR4 (~100 kDa), preferentially recognizing the TLR4/MD-2 complex. It has been validated for use in flow cytometric analysis of mouse bone marrow-derived dendritic cells at ≤0.5 µg/test (100 µL final volume).
Careful titration is recommended for optimal assay performance.

Super Bright 436 can be excited with a violet laser (405 nm) and emits at 436 nm. Use a 450/50 bandpass filter or equivalent.
When using multiple Super Bright dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.

Target Information

MD-2 is part of the Toll/interleukin-1 receptor (TIR) family, including TLRs that recognize pathogen-associated molecular patterns (PAMPs) and mediate innate and adaptive immune responses.
TLR4, the major signaling receptor for LPS, requires MD-2 binding for maximal response. The species of MD-2 determines response specificity—for instance, human MD-2 expressed in mouse cells alters TLR4 responsiveness to LPS analogs.
