Target Information
STAT3는 STAT (signal transducers and activators of transcription) 단백질 계열에 속하며, 수용체 관련 키나아제에 의해 인산화되어 핵으로 이동 후 전사 인자로 작용합니다.
STAT3는 Interferon-alpha, Interferon-gamma, EGF, PDGF, IL6 등에 의해 활성화될 수 있으며, JAK1 및 JAK2에 의해 Tyr705 잔기에서 인산화되어 이합체 형성, 핵 내 이동, DNA 결합에 필수적입니다.
인간 STAT3 유전자는 염색체 17의 q arm에 위치하며, IFN-alpha에 의해 활성화되지만 IFN-beta에는 반응하지 않습니다.
STAT3는 다양한 유전자 발현을 매개하여 세포 성장 및 세포사멸 등 여러 세포 과정에서 핵심 역할을 합니다.
Rac1 GTPase 및 PIAS3 단백질은 각각 STAT3의 활성 조절 및 억제에 관여합니다.
STAT3 유전자는 세 가지 대체 스플라이싱 변이체를 가지며, knockout 마우스에서의 결실은 배아 초기 단계에서 치명적입니다.
