Target Information
Estrogen Receptors (ER) are members of the steroid/thyroid hormone receptor superfamily of nuclear receptors. The estrogen receptor is a ligand-activated transcription factor that, when bound to estrogen, undergoes a conformational change enabling dimerization and binding to estrogen response elements (ERE) in DNA. This binding regulates gene transcription through coactivator or corepressor recruitment.
There are two forms of the estrogen receptor, alpha and beta, encoded by ESR1 and ESR2 genes. Alternative RNA splicing produces at least four estrogen receptor-alpha isoforms (66 kDa, 53 kDa, 47 kDa, 35 kDa). ERs are widely expressed across tissues, essential for sexual development and reproductive function, and involved in bone metabolism. They are implicated in diseases such as breast cancer, endometrial cancer, and osteoporosis.
