Enolase (2-phosphoglycerate hydrolyase or phosphopyruvate hydrates) is a glycolytic enzyme that catalyzes the conversion of 2-phosphoglycerate to phosphoenolpyruvate. It consists of three subunits: alpha, beta, and gamma.
Non-neuronal enolase (ENO1) is an isoform composed of two alpha subunits and also encodes a shorter monomeric structural lens protein, tau-crystallin.

This multifunctional enzyme is involved in glycolysis, growth control, hypoxia tolerance, and allergic responses. It may also act as a receptor and activator of plasminogen on the cell surface of various cell types, including leukocytes and neurons, and can stimulate immunoglobulin production.

ENO1 is used as a diagnostic marker for many tumors and, in its alpha/gamma heterodimeric form, serves as a marker for hypoxic brain injury after cardiac arrest. It is also a marker for endometriosis. Antibodies against alpha-enolase are found in sera from patients with cancer-associated retinopathy syndrome (CAR) and have been identified as autoantigens in Hashimoto encephalopathy (HE), associated with Hashimoto thyroiditis (HT).
