Target Information
Importin 9 functions in nuclear protein import as a nuclear transport receptor. It serves as a receptor for nuclear localization signals (NLS) in cargo substrates and mediates docking of the importin/substrate complex to the nuclear pore complex (NPC). The complex is translocated through the pore by an energy-requiring, Ran-dependent mechanism.
At the nucleoplasmic side, Ran binds to importin, causing dissociation of the complex, and importin is re-exported to the cytoplasm where GTP hydrolysis releases Ran.
This mechanism confers directionality of nuclear import through asymmetric distribution of Ran-GTP and Ran-GDP forms.
Importin 9 mediates nuclear import of H2B histone, RPS7, and RPL18A, preventing cytoplasmic aggregation of RPS7 and RPL18A. It may also import H2A, H3, H4 histones, RPL4, and RPL6.
