Description:
The PC61.5 antibody reacts with mouse CD25, the 55 kDa interleukin-2 receptor alpha chain (IL-2Rα). CD25 is expressed by early progenitors of T and B lineages and by activated mature T and B lymphocytes. By itself, CD25 binds IL-2 with low affinity, but forms a high-affinity receptor when associated with CD122 (IL-2Rβ) and CD132 (common γ chain). The PC61.5 antibody inhibits IL-2 binding to both receptor classes, playing a role in lymphocyte differentiation and activation/proliferation.

This antibody is reported for use in flow cytometric analysis.

Applications Tested:
Tested by flow cytometric analysis of mouse splenocytes. Recommended use ≤0.25 µg per test (100 µL final volume). Cell numbers from 10⁵–10⁸ cells/test; titration recommended for optimal performance.

Target Information

CD25 (IL2 receptor alpha chain, IL2RA) is a cytokine receptor involved in T and B lymphocyte proliferation. The IL2RA forms a high-affinity receptor complex with IL2RB and IL2RG. Homodimeric IL2RA yields low-affinity receptors, while IL2RB homodimers yield medium affinity. IL2 expression in mature thymocytes is monoallelic, regulating precise gene expression. IL2 primarily produced by T cells acts as a growth, differentiation, and survival factor. It stimulates B cell proliferation, NK cell activity, and inhibits granulocyte macrophage colony formation. Disruption of the IL2RA gene in mice causes ulcerative colitis-like disease, indicating its role in immune response regulation. Mutations are associated with IL2 receptor alpha deficiency.
