Casein kinase I (CKI) and casein kinase II (CKII) are families of serine/threonine protein kinases found in all eukaryotes. CKI family members, including CKI alpha, gamma, epsilon, and delta, regulate cytoplasmic and nuclear processes such as DNA replication and repair. CKII typically forms a tetrameric complex (alpha2 beta2 or alpha alpha' beta2), with the alpha catalytic subunit activated by the beta regulatory subunit. CKII is abundant in proliferating and differentiating cells, phosphorylating over 100 substrates including nuclear oncoproteins, transcription factors, and DNA metabolism enzymes.
