Target Information

CD30 (Ki-1, TNF Receptor Superfamily Member 8) is a type I transmembrane glycoprotein belonging to the TNF receptor superfamily. It was originally identified as a cell surface antigen of Hodgkin’s and Reed-Sternberg cells using monoclonal antibody Ki-1. The ligand for CD30 is CD30L (CD153), and their interaction mediates various effects including cell proliferation, activation, differentiation, and apoptosis.

CD30 plays a critical role in the pathophysiology of Hodgkin’s disease and other CD30⁺ lymphomas. It functions as a costimulatory molecule in thymic negative selection and is expressed in Hodgkin’s and Reed-Sternberg cells, some non-Hodgkin’s lymphomas, virus-infected T and B cells, and activated normal T and B cells. In T cells, CD30 is found on subsets producing Th2-type cytokines and on CD4⁺/CD8⁺ thymocytes co-expressing CD45RO and IL4 receptor.

The soluble form of CD30 (sCD30) serves as a marker of Th2 immune response. CD30 interacts with TRAF2 and TRAF5, mediating NF-κB activation and regulating apoptosis. It limits the proliferation of autoreactive CD8 effector T cells, contributing to immune homeostasis. Two alternatively spliced transcript variants encoding distinct isoforms have been reported.

CD30 expression is characteristic of mononuclear cells in Hodgkin’s lymphoma, Reed-Sternberg cells, most Anaplastic Large Cell Lymphomas (ALCL), and embryonal carcinomas.
