Target Information

Protein disulfide isomerase (PDI) is a multifunctional protein that catalyzes the formation, breakage, and rearrangement of disulfide bonds. At the cell surface, it acts as a reductase that cleaves disulfide bonds of proteins attached to the cell, potentially causing structural modifications. Inside the cell, PDI forms and rearranges disulfide bonds of nascent proteins.
At high concentrations, it functions as a chaperone preventing aggregation of misfolded proteins, while at low concentrations, it facilitates aggregation (anti-chaperone activity). PDI may cooperate with other chaperones in structural modification of TG precursor during hormone biogenesis. It also serves as a structural subunit of enzymes such as prolyl 4-hydroxylase and microsomal triacylglycerol transfer protein (MTTP).
It acts as a receptor for LGALS9, retaining P4HB at the surface of Th2 T helper cells, increasing disulfide reductase activity at the plasma membrane, altering redox state, and enhancing cell migration.
