Target Information
IL-27, a member of the IL-12 family, is a heterodimeric protein consisting of the p40-related protein Epstein-Barr virus-induced gene 3 (EBI3) non-covalently linked to an IL-12p35-related protein, p28 (also known as IL-30). IL-27 is produced by activated APCs and mature dendritic cells.
IL-27 acts on NK cells and naive CD4+ T cells, and receptor expression analysis (WSX-1/TCCR and gp130) indicates possible action on mast cells and monocytes. Binding of IL-27 to WSX-1/gp130 activates JAK1, STAT1, and STAT3, leading to STAT1/3 phosphorylation.
WSX-1/TCCR-deficient mice show impaired Th1 responses and increased susceptibility to L. monocytogenes infection, suggesting IL-27’s role in Th1 response initiation. IL-27 induces T-bet, a Th1-specific transcription factor, and suppresses the Th2-specific factor GATA-3. This regulation supports Th1 differentiation and IL-12Rβ2 expression maintenance in activated CD4+ T cells.
IL-27 also exhibits antitumor activity by enhancing CTL function, granzyme B expression, and promoting tumor regression in vivo.
