Transforming Growth Factor (TGF) betas mediate many cell-to-cell interactions that occur during embryonic development. Three TGF betas have been identified in mammals: TGF beta 1, TGF beta 2, and TGF beta 3. Each is synthesized as a precursor protein that is cleaved to yield a 112 amino acid polypeptide associated with its latent portion.

TGF beta polypeptides are multifunctional, influencing cell proliferation, differentiation, and other cellular functions. Both transformed and non-neoplastic tissues release TGF betas, and most mammalian cells possess specific TGF receptors.

TGF beta exhibits multimodal activity, capable of stimulating or inhibiting cellular proliferation. Generally, mesenchymal cells are stimulated, while epithelial or neuroectodermal cells are inhibited. TGF beta 1, beta 2, and beta 1.2 share similar biological activity but may differ in receptor binding.

TGF beta 2 is produced by many cell types, found in highest concentrations in porcine platelets and mammalian bone. Latent TGF beta 2 is the dominant isoform in body fluids such as amniotic fluid, breast milk, and ocular humors.
