The EMK08 monoclonal antibody reacts with human CD133 (Prominin-1), a 120 kDa pentaspan membrane protein also known as Prominin-2 family member. CD133 is expressed on plasma membrane protrusions such as microvilli and exists in multiple isoforms with tissue-dependent glycosylation patterns. Initially identified on CD34+ hematopoietic progenitors, CD133 is widely used as a marker for stem cells and cancer stem cells, including colon, brain, and prostate cancer stem cells.

The EMK08 antibody binds a distinct epitope from TMP4 (Product #12-1338) and does not interfere with TMP4 binding.

Target Information

CD133 (AC133) antigen is a 97 kDa glycoprotein with five transmembrane domains, appearing as a 120 kDa band due to glycosylation. It is a marker for immature hematopoietic stem cells and various stem/progenitor cells such as neural and embryonic stem cells. CD133 is also expressed in certain cancers, including leukemias and brain tumors, and at low levels in kidney, pancreas, placenta, and fetal liver.

Mutations in CD133 are associated with retinitis pigmentosa and Stargardt disease. Expression is regulated by at least five alternative promoters, producing multiple isoforms depending on tissue type.
