This 15-414 monoclonal antibody reacts with human CD172a (signal regulatory protein alpha, SIRP alpha).
CD172a is a cell surface glycoprotein with three Ig-like extracellular domains and two cytoplasmic ITIM motifs. These ITIMs recruit SHP-1 and SHP-2 phosphatases.
CD172a is expressed on monocytes, granulocytes, dendritic cells, and CD34+CD38− progenitor cells, but not on T or B lymphocytes. Neurons also express CD172a.
Its extracellular ligand is CD47, and it plays roles in dendritic cell-mediated T cell activation, neutrophil migration, and phagocytosis.

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and validated for flow cytometry using human peripheral blood cells at 5 µL (0.5 µg) per test.

Super Bright 600 is a tandem dye excited by a violet laser (405 nm) and emits at 600 nm. Recommended filter: 610/20 bandpass.

Staining Recommendations:

• Use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.
• Protect from light due to photo-induced oxidation sensitivity.
• Samples may be fixed using IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333) and stored up to 3 days at 4°C in the dark.

Target Information

SIRP alpha (CD172a) is a receptor-type transmembrane glycoprotein expressed on myeloid cells such as granulocytes, dendritic cells, macrophages, mast cells, and hematopoietic stem cells.
It acts as a substrate for tyrosine kinases (EGFR, PDGFR, src, insulin receptor) and negatively regulates receptor tyrosine kinase-coupled signaling pathways.
Binding to CD47 leads to phosphorylation of ITIMs, recruiting SHP-1 and SHP-2 phosphatases, and modulates T cell and dendritic cell activation.
SIRP alpha influences cellular responses to growth factors, hormones, and adhesion, and plays roles in physiological and pathological processes.
Cancer cells expressing CD47 activate SIRP alpha, inhibiting macrophage-mediated phagocytosis of tumor cells.
