Target Information

Mammalian apurinic/apyrimidinic endonuclease (APE/ref-1) is a multifunctional enzyme crucial for DNA repair and redox regulation.
APE repairs abasic sites in DNA and modulates the redox state of proteins involved in oxidative signaling and transcription factor regulation (Fos, Jun, NF-kB, Myb, HIF-1α, CREB, Pax), cell cycle control (p53), and apoptosis.
Abasic sites arise from spontaneous loss or oxidative damage, chemotherapy, ionizing radiation, UV irradiation, and DNA glycosylase activity.
APE expression varies by tissue and developmental stage, showing diverse subcellular localization and interactions.
Regulation by phosphorylation occurs partly through casein kinase II.
Increased APE levels are observed upon reoxygenation of hypoxic cells and in some malignancies, while decreased expression is linked to apoptosis induction.
