Target Information

Nucleocytoplasmic transport is a signal- and energy-dependent process occurring through nuclear pore complexes in the nuclear envelope. Proteins containing a nuclear localization signal (NLS) are imported via a receptor complex composed of importin alpha and beta (karyopherins). Importin alpha binds NLS-containing cargo, while importin beta docks the complex to the nuclear pore. In the presence of nucleoside triphosphates and Ran GTPase, the complex translocates through the pore, after which importin subunits dissociate. Importin beta remains at the pore, while importin alpha and the cargo enter the nucleoplasm. Interactions between importin beta and FG repeats of nucleoporins are essential for translocation.
