Target Information
Tight junctions form an important barrier for paracellular transport in epithelial cells. Tricellulin is the first protein identified that specifically concentrates in tricellular tight junctions, where three adjacent cells meet. It has four membrane-spanning domains similar to claudins and is highly expressed in epithelial tissues such as the small intestine, kidney, and lung. Functional studies using siRNA have shown that tricellulin is essential for maintaining epithelial barrier integrity and tight junction formation. Mutations affecting its cytosolic domain, which interacts with the scaffolding protein ZO-1, lead to impaired tight junction function.
